Understanding the mechanism of iPS cell induction through the study of the primordial germ cells

Primordial germ cells (PGCs) potentially maintain pluripotency during embryogenesis, yet the underlying mechanism remains unknown. First, I plan to establish the robust strategy for reconstituting the mammalian germ cell lineage from ESCs and iPSCs in vitro. Together, these technological innovations will make it possible to analyze genetic and epigenetic molecular mechanisms that underlie the germ cell proper via both biochemical and cellular functional approaches. Through this study of the maintenance of PGC potential pluripotency, I will seek a better understanding of the induction of the iPSCs.
