Abstract
RNAi is a form of sequence-specific gene silencing induced by the introduction of double-stranded RNA, and is a useful tool for regulating gene expression in eukaryotic cells. Based on the accumulated knowledge obtained from genetic studies in the nematode C. elegans, I will analyze the mechanism of RNAi with genetic as well as biochemical approaches. A cell-free reaction system have been developed and will be utilized for the biochemical analysis. In order to explore conditions enhancing the RNAi response in organisms, analyses will be focused mainly on the downstream reaction and the signal amplification in the RNAi pathway.
